Born at Besthorpe, Norfolk, 21 Jul 1597, and he was baptized at Besthorpe on 26 Jul 1597. He died at Westminster, Middx., 8 Nov 1639, and he was buried at Besthorpe on 15 Nov 1639. He married Mary Cokayne at St. Peter le Poer, City of London, on 26 Nov 1633. Mary Cokayne, the daughter of William Cokayne and Eleanor Lloyd, was baptized at St. Peter le Poer, City of London, on 22 Jul 1610, and she was buried at Besthorpe on 20 Jun 1688. Knighted 1638. Sir William Drury and Mary Cokayne had five children:

William, who was baptized at St. Peter le Poer, City of London, 6 Oct 1634, and buried at St. Peter le Poer, City of London, 8 Oct 1634.

Bridget, who was baptized at Besthorpe 21 Jan 1635/36, and who married first Charles Needham, 4th Viscount Kilmorey, second, Sir John Shaw, and third Sir John Baber.

Jane, who was baptized at Besthorpe 1 Apr 1638, and buried at Besthorpe 12 Jul 1638.

Anthony, who was baptized at Besthorpe 17 Feb 1638/39, and buried at Besthorpe 14 Sep 1640.

Anne, who was baptized at Besthorpe 12 Jul 1640, and buried at Besthorpe 12 Jan 1678/79. At her baptism, it was recorded that William Drury, her father, was dead. Anne married Phillip Harbord of Stanninghall, Norfolk, who was buried at Besthorpe 13 Sep 1687.

A Monumental Inscription in the chancel of All Saints, Besthorpe, refers to the burial. Sir William Drury's Will, dated 7 Nov 1639, and probated 20 Jan 1639/40, mentioned his brother, Anthony, and Anthony's oldest dau. (unnamed), his four sisters (unnamed), his dau., Bridget, his eldest son (unnamed), his child not yet born, and his wife, Mary, as his sole executrix, the overseers being his uncle, William Drury, his brother-in-law, Robert Wilton, and his father-in-law, William Cokayne, and the witnesses being Francis Vernon, James Tyrell, Thomas Cokayne, and Francis Mosse.
